Role of Hsp70 Chaperone HSPA1L in Protein Folding and Regulation of Glucocorticoid ReceptorProject is carried out within the SONATA 20 grant financed by National Science Centre, Poland |
Project description: |
Hsp70 chaperones play a critical role in cell protection against protein damage, determining cell fate during stress, aging and proteotoxic diseases. Hsp70 is a central hub of a network of chaperone systems that collectively support protein folding, prevent aggregation of misfolded proteins, recover active proteins from polypeptides trapped in aggregates and regulate many proteins in their native, physiological state. One of the Hsp70 family members is HSPA1L protein. Recent studies show significant association of mutations and polymorphisms of the HSPA1L gene with several prevalent conditions with an autoimmunological and inflammatory component. The project will involve generation and purification of chaperones, their variants and protein substrates. Subsequently, using biochemical analyses and, in parallel, reporter cell lines, we will inquire into the effects of the disease-linked substitutions in HSPA1L on its chaperone activity, with particular emphasis on chaperone- dependent regulatory cycle of Glucocorticoid receptor. The research aims to unravel novel mechanisms of the interplay between the Hsp70s in health and diseases involving dysfunctional glucocorticoid signalling. |
