Role of Hsp70 Chaperone HSPA1L in Protein Folding and Regulation of Glucocorticoid Receptor

Project is carried out within the SONATA 20 grant financed by National Science Centre, Poland

Project description:

Hsp70 chaperones play a critical role in cell protection against protein damage, determining cell fate during stress, aging and proteotoxic diseases. Hsp70 is a central hub of a network of chaperone systems that collectively support protein folding, prevent aggregation of misfolded proteins, recover active proteins from polypeptides trapped in aggregates and regulate many proteins in their native, physiological state. One of the Hsp70 family members is HSPA1L protein. Recent studies show significant association of mutations and polymorphisms of the HSPA1L gene with several prevalent conditions with an autoimmunological and inflammatory component. The project will involve generation and purification of chaperones, their variants and protein substrates. Subsequently, using biochemical analyses and, in parallel, reporter cell lines, we will inquire into the effects of the disease-linked substitutions in HSPA1L on its chaperone activity, with particular emphasis on chaperone- dependent regulatory cycle of Glucocorticoid receptor. The research aims to unravel novel mechanisms of the interplay between the Hsp70s in health and diseases involving dysfunctional glucocorticoid signalling.

Key responsibilities include:

  1. performing research using biochemical, genetic and molecular biology techniques (independently and in collaboration with other team members)
  2. participation in design of experiments and data analysis
  3. participation in preparation and writing of manuscripts
  4. supervising younger researchers (PhD students and Master students) involved
    in the project
  5. active participation in lab meetings, departmental seminars and conferences

Profile of candidates/requirements:

  1. Doctoral degree in biochemistry, molecular biology or a similar discipline obtained in 2013 or later (candidates who do not hold a doctoral degree may apply for the position — in this case, signing the contract will be conditional upon submitting a document confirming the attainment of the doctoral degree)
  2. Publication record and experimental training in biochemistry and molecular biology. Publication(s) in the field of protein quality control will be an asset.
  3. Fluent English language and good communication skills
  4. Strong motivation to pursue scientific questions, creativity and analytical thinking
  5. Research experience in protein purification, protein-protein interactions analysis, enzymatic activities analysis and genetics.
  6. The experience in human cell culture will be advantageous
